Clinical Significance
NRAS, a member of RAS family with gtpase activity, is a key regulator of cell growth, proliferation and differentiation. Activating mutations of NRAS have been found in a variety of cancers, including melanoma (15-25%) and colorectal cancer (2-6%).
The common mutation sites of NRAS gene located in exon 2, 3 and 4, mainly in codon 12, 13, 59, 61, 117 and 146. Some studies have found that the mutation frequency of NRAS gene exon 2 (codons 12, 13) in colorectal cancer patients with wild-type KRAS gene exon 2 is 3.8%, exon 3 (codons 59, 61) is 4.8%, and exon 4 (codons 117, 146) is 0.5%.


Clinical studies have confirmed that detection of NRAS gene mutation status can predict the treatment response rate of cetuximab in patients with metastatic colorectal cancer.
